about 2 weeks into it (not even taking it everyday) and the headache is gone, I am loving this drug, I don´t know if it´s placebo effect or what but it is giving me some extra sexual performance and confidence, also reducing refractory period.
It is a really interesting drug:
Research on the effects of tadalafil, a phosphodiesterase type-5 (PDE5) inhibitor, on hormones indicates several significant impacts, particularly concerning sex steroid hormones and related pathways.
- Testosterone and Estradiol Levels: Tadalafil has been shown to increase total and free testosterone levels while decreasing estradiol levels in studies involving both human subjects and animal models. This modulation is hypothesized to occur through the inhibition of aromatase activity, which is responsible for converting testosterone to estradiol. This effect can be beneficial in conditions like erectile dysfunction and potentially in the management of prostate health (BMJ Diabetes Research & Care) (MDPI).
- Androgen Receptor Activity: Tadalafil has been found to increase the expression and activity of androgen receptors (AR). This was demonstrated in cellular models where tadalafil treatment enhanced AR protein expression and promoted AR nuclear translocation and transcriptional activity. This indicates that tadalafil might support androgenic actions in tissues beyond its primary use for erectile dysfunction (SpringerLink) (MDPI).
- Bone and Adipose Tissue: In studies involving bone and adipose tissues, tadalafil has shown potential in modulating hormone pathways that could influence bone density and adipose tissue function. It may reduce the expression of aromatase in osteoblasts, which could impact bone health positively by maintaining higher levels of testosterone and lower levels of estradiol (MDPI) (SpringerLink).
- Inflammatory and Metabolic Parameters: Research involving diabetic rats treated with tadalafil revealed improvements in lipid profiles, including reductions in LDL cholesterol and triglycerides. These changes were accompanied by increased testosterone levels and improved copulatory behavior, suggesting broader systemic effects of tadalafil on metabolism and hormone balance (BMJ Diabetes Research & Care).
