I would appreciate it if you all could look at my hypothetical proposed research project for enhancing hefer muscle growth.
I have outlined my proposed process that I came up with using your posts. If you have any corrections or additions please feel free to voice them.
Mortar and Pestle, Non-Porous
Cottonseed Oil, USP
Nalgene 22um vial top filter // or cell culture bottle top filter., PVDF Membrane
Finaplex-H
10ml serological pipette, sterile
Laminar Flow Hood
1) Grind 6g of pellets into fine powder using mortar, 400 mesh size.
2) Completely dissolve powder into oil (100mL)
3) Autoclave solution using dry cycle.
4) Filter solution using Nalgene bottle top cell culture filter.
5) Transfer using aseptic technique into individual rubber top vials.
OR
4) Filter into cell culture bulk 22um filter.
5) Pipette into 10mL rubber top bottles, crimp.
6) Sterilize with EO or Gamma irradiation.
Is there really a need to autoclave/sterilize if using the cell culture filter?
We could plate out a sample of the end product onto some TSA media to check for bacterial growth... I would also HPLC the sample to check for impurities.
